Protein Purification A company was unable to achieve effective purification of a potential therapeutic protein from a high level bio-production system. ABT scientists determined that the purification problems were due to reversible aggregation of the protein of interest with host proteins. ABT was able to devise conditions that effectively disrupted the aggregate and allowed efficient purification.
Antibody Stabilization Solution formulations were developed by ABT that effectively stabilize monoclonal antibodies to high temperatures that may occur during shipping in the absence of temperature controls. This technology also has applications for delivering diagnostics to remote areas of the third world. Antibody Expression and Characterization ABT is working with a client to develop a prototype bioproduction method for generating therapeutic antibodies with improved pharmacological properties. The antibodies produced by this method are characterized by determination of their antigen dissociation constants, in solution, as well as by gel electrophoresis, Western blotting, and size exclusion chromatography.
Solubilization of a Protein Therapeutic in Ethanol A client requested that ABT determine whether any current protein therapeutic other than insulin might be compatible with a deliver system that required the protein to be soluble in ethanol. ABT selected a protein therapeutic and was able to solubilize it in 100% ethanol with complete retention of antibody binding activity.
Protein Labeling A company that produces diagnostic kits had monoclonal antibodies in their inventory that possessed useful antigen binding properties but that lost all binding activity after fluorescent labeling with FITC. ABT devised a novel, alternative labeling scheme that resulted in active labeled antibodies. These labeled antibodies are currently being formulated for use in test kits.
